The role of octamer binding transcription factors in glioblastoma multiforme

A group of transcription factors (TF) that are master developmental regulators of the establishment and maintenance of pluripotency during embryogenesis play additional roles to control tissue homeostasis and regeneration in adults. Among these TFs, members of the Octamer-binding transcription factor (OCT) gene family are well documented as major regulators controlling the self-renewal and pluripotency of stem cells isolated from different adult organs including the brain. In the last few years a large number of studies show the aberrant expression and dysfunction of OCT in different types of cancers including glioblastoma multiforme (GBM). GBM is the most common malignant primary brain tumor, and contains a subpopulation of undifferentiated stem cells (GSCs), with self-renewal and tumorigenic potential that contribute to tumor initiation, invasion, recurrence, and therapeutic resistance. In this review, we have summarized the current knowledge about OCT family in GBM and their crucial role in the initiation, maintenance and drug resistance properties of GSCs. Background: Obstructive sleep apnea (OSA) is a common disorder of repetitive pharyngeal collapse during sleep. It is more common among middle-aged adults. Obesity leads to and intensifies OSA. Epidemiologic studies have recorded relations of OSA with hypertension. The Berlin Questionnaire may find persons at high possibility for OSA and hence can evade costly polysomnography (PSG) studies, especially in resource-limited settings. This study is about our experience of Berlin Questionnaire and clinical characteristics of patients with OSA. Objective: To reveal our experience of OSA in terms of (i) age/sex distribution; (ii) Berlin Questionnaire utility; (iii) clinical features; (iv) polysomnograhic findings; and (v) association with obesity and hypertension. Materials and Methods: This prospective observational study was done in the Department of Chest Medicine and Department of ENT, Head and Neck Surgery of Government Medical College, Srinagar, Jammu and Kashmir. Outpatient department/inpatient department patients who showed positive scoring on Berlin Questionnaire were enrolled in the study. PSG was done in all of these patients. Clinical features, blood pressure, and body mass index (BMI) were noted in patients who showed positive PSG. Result: This study included a total of 120 Berlin Questionnaire positive patients, of which 96 were positive on PSG. Positive predictability value of Berlin Questionnaire was 80% for both low- and high-risk cases. Majority of PSG positive cases were female subjects (68.75%) while males comprised 31.25% of PSG positive cases. Most common age group was 5165 years. Of PSG positive OSA patients, 41 (42.7%) showed mild disease, 30 (31.2%) moderate disease, and 25 (26%) severe disease. Most patients were grouped as per BMI into obese class II, and majorities were subjects with hypertension. In addition to snoring, other clinical features were dry mouth, excessive daytime sleepiness, and irritability. Conclusion: Berlin Questionnaire is an important screening tool to detect suspected OSA patients. Although male subjects are more commonly seen to be affected in literature, we found female predominance. Most of these patients are middle-aged. Obesity is an important risk factor, and most of these patients show BMI > 30 kg/m2. Most of these patients present hypertension as a comorbidity.

Background: Inappropriate feeding practices including breast-feeding practice during the first year of life identified to be associated with 60% of child malnutrition and mortality. Lack of feeding skills is the major cause. World Health Organization (WHO) mentions that growth chart is a good educational tool for parents. It needs an innovation to introduce growth chart. Objective: The objective of this study is to assess the effect of scores in measuring child growth status on feeding practices and calorie intake. The hypothesis of this study is that after intervention there would be a significant improvement of feeding practices which in turn will affect the calorie intake of children in the intervention group. Materials and Methods: This study is to measure the impact of using score bubble growth chart on infant feeding practices and calorie intake. A total of 107 caregivers selected purposively from two different locations. Mothers in intervention group were trained to use score bubble growth chart while mothers in control group used the normal growth chart. Sixteen written questions were designed and tested to measure mothers behavior in feeding their babies. Data were analyzed using independent t-test and chi-square test. Result: Before intervention, there was no significant difference of infant feeding practices and types of food given (p > 0.05) and child calorie intake (p > 0.05) between intervention and control groups. Understanding the occupational traveller profile towards malaria prevention enables directed health promotion and preventive practices

Aim/background: Increased migration and expatriation to malaria endemic countries has been appointed as contributing factor to the increasing imported malaria hospitalizations recently observed in Portugal. This study sought to profile this traveller and understand the knowledge, attitude and practice (KAP) towards malaria prevention as necessary first step to improve and suit risk reduction strategies. Methods: A cross sectional questionnaire based study was conducted at the Lisbon International Airport aiming to characterize the Portuguese occupational traveller to Angola. Travel and travellers characteristics associated to not seeking travel health advice and KAP towards malaria were specifically studied. Results: Occupational travellers represented 92,6% (n= 374/404) of the Portuguese travellers to Angola that participated in the survey. The profile found was that of an occupational traveller experienced in developing destinations to urban settings for long periods. The knowledge regarding malaria risk and prevention was good but the same was not observed for preventive practices. Those with chronic health conditions and those living in Angola or both in Portugal and Angola tend not to seek pre-travel medical advice and to be at higher malaria risk, considering their KAP towards malaria. Conclusions: Increasing watchfulness concerning occupational travellers and expatriates need to be in the public health agenda and suited interventions are needed.

The Reality of Medical Services Provided to the People with Special Needs in the Majmaah City from the Viewpoint of Specialists

Physiotherapy, occupational therapy, nursing and support services are necessary services provided to people with special needs. Purpose: the aim of this study is to emphasize the reality of medical services provided to the people with special needs in the Majmaah city from the viewpoint of various specialists Materials and Methods: This is a Descriptive study. A Structured self-Administered questionnaire was completed by the specialists in different fields in the hospital to assess medical services provided to the people with special needs at the health centers in Majmaah province, Saudi Arabia Results: The study was consisted of five questions in the questionnaire. The first question showed that the level of medical services provided to the disabled in the province from the viewpoint of specialists, the second question also showed statistically significant difference at the level of (α = 0.05) medical services provided to the disabled in the province from the viewpoint of specialists such as doctors and nurses, the third question showed that differences in the level of medical services provided to the disabled in the province from the viewpoint of specialists according to the qualification is not a statistical function at a level of significance (α = 0.05) the fourth question also showed differences in the level of medical services provided to the disabled in the province from the viewpoint of specialists according to expertise not statistically function at a level of significance (α = 0.05) finally the fifth question, the answer was that the differences in the level of medical services provided to the disabled in the province from the viewpoint of specialists according to the training courses non-statistically function at a level of significance (α = 0.05) Conclusion: This study concludes that the level/quality of medical services provided to the disabled people in Majmaah province need to be improved and they must have a consistency in the delivery of medical services.

Previously we established Zygote Electroporation of Nucleases (ZEN) technology as an efficient and high-throughput way to generate genetically modified mouse models. However, there were significant variations of the targeting efficiency among different genomic loci using our previously published protocol. In this study, we improved the ZEN technology by delivering Cas9 protein into mouse zygotes through a series of electroporation. Using this approach, we were able to introduce precise nucleotide substitutions, large segment deletion and short segment insertion into targeted loci with high efficiency.

Cardiorespiratory fitness and cognitive functioning following short-term interventions in chronic stroke survivors with cognitive impairment: a pilot study.

This study, a quasi-experimental, one-group pretest-post-test design, evaluated the effects on cognitive functioning and cardiorespiratory fitness of 8-week interventions (aerobic exercise alone and aerobic exercise and cognitive training combined) in patients with chronic stroke and cognitive impairment living in the community (participants: n=14, 61.93+/-9.90 years old, 51.50+/-38.22 months after stroke, n=7 per intervention group). Cognitive functions and cardiorespiratory fitness were evaluated before and after intervention, and at a 3-month follow-up visit (episodic memory: revised-Hopkins Verbal Learning Test; working memory: Brown-Peterson paradigm; attention omission and commission errors: Continuous Performance Test; cardiorespiratory fitness: peak oxygen uptake during a symptom-limited, graded exercise test performed on a semirecumbent ergometer). Friedman's two-way analysis of variance by ranks evaluated differences in score distributions related to time (for the two groups combined). Post-hoc testing was adjusted for multiple comparisons. Compared with before the intervention, there was a significant reduction in attention errors immediately following the intervention (omission errors: 14.6+/-21.5 vs. 8+/-13.9, P=0.01; commission errors: 16.4+/-6.3 vs. 10.9+/-7.2, P=0.04), and in part at follow-up (omission errors on follow-up: 3.4+/-4.3, P=0.03; commission errors on follow-up: 13.2+/-7.6, P=0.42). These results suggest that attention may improve in chronic stroke survivors with cognitive impairment following short-term training that includes an aerobic component, without a change in cardiorespiratory fitness. The psychometric properties of the Turkish Stroke and Aphasia Quality Of Life Scale-39.

The Stroke and Aphasia Quality Of Life Scale (SAQOL-39) is a widely used instrument in assessing the quality of life in aphasic patients. Our purpose was to translate the SAQOL-39 into the Turkish language (SAQOL-39/TR) and assess its reliability and validity in patients who had aphasia. SAQOL-39/TR was obtained using the 'translation-backward translation' method and administered to 40 patients with aphasia. The reliability studies were performed by means of internal consistency and test-retest reliability. The validation studies were carried out by means of construct validity using within-scale analyses and analyses against the external criteria. Correlation analysis was performed between scales and the Ege Aphasia Test, the Barthel index, the 12-item General Health Questionnaire and the Brunnstrom recovery stages (BRS) of the arm, hand, and lower extremity. In the results, the scores of the SAQOL-39 were not different between groups. Cronbach's [alpha] variables were good for all domains (0.80, 0.88, 0.89, 0.82, and 0.83). Test-retest reliability was also high (0.96, 0.97, 0.91, 0.70, and 0.96). There were significant correlations with coefficients ranging from 0.36 to 0.60 among the domains of scale and other measures. Moderate-high correlations were also seen with BRS-arm, BRS-hand, and BRS-lower extremity (r, 0.27-0.58).
